Hughes is a faculty member and coordinator of the secondary education program in the School of Education, Howard University. She is a co-principal investigator of the Center for Research on the Education of Students Placed at Risk classroom assessment project, an interdisciplinary research project examining the problems of effective classroom assessment in mathematics and reading. This project, and other work, reflects her contributions to understanding the best ways to judge the educational achievement of minority children. Hughes' work has been primarily in educational measurement, including serving on the National Assessment of Educational Progress (NAEP) validation studies panel as a mathematics and test- and item-bias consultant. She has a strong commitment to mentoring and affirmative action and is a member of the American Educational Research Association's Division D Affirmative Action Committee.

Her work in evaluation has been influenced by Floraline Stevens and Stafford Hood, as well as Edmund Gordon and, especially, Sylvia T. Johnson, who was known as a crusader for quality and fair testing for minorities.
